DNA metabarcoding can be applied as a quick screening of the bentic biodiversity in a large number of samples. Certain protocol choices could however influence the detection success.

 

DNA extraction of the mixed sample still yields better results than just using the ethanol in which the sample was stored. The accurate identification of a species is dependent on its size and the presence of chitine or CaCO3.
